It could have some impact on temperature if you take the temp at the top of the water!
Also, certain tests are dependent on the time of day-as differing results will be evident from that of day time and nighttime, such as pH, plus it is best to test at the same time of the day for most accurate results!

With good water movement in the tank a sample from the top, middle, bottom, back, front, left or right should be the same in terms of make-up, temp and clarity.

    JMHO :)

Hey! The spot doesn't matter! The only thing that matter is the fact you should repeat the test at least 2 times with 5 min interval! Many people do not know it and/or forget about it! Simple aquaristic test kits are highly inaccurate, especially Ca2+/Ma2+ test! Moreover, these are colorimetric test, that are strongly biased by your perception; e.g. ambient light in which you read the outcome of the test!

    thanks,
i just figuerd that since most of the waste gathers at the top, this is why skimmers have a overflow. so i thought maybe water parims would be slightly inaccurate, if i took water from the surface.
oh yeah i have no sump, so my test would be out of the main display.

pH will be different in the sump than the main tank
