In the near future I hope to start a new build and would like to try drilling a tank for the first time (only tempered on the bottom). Im still on the fence as to the exact method of overflow I use, but my main question is, when working with acrylic, say.... building a DIY internal overflow box..... what is the best/safest, glue/adhesive/epoxy... etc.

Thank you for the suggestions. I didn't plan on using silicone for assembly of the box, only to adhere the box to the glass. now I just have to decide on design.
